In this thesis, the dental plaque-pH after acid and alkali formation in vivo in relation to the individual caries status were examined. Two chair-side methods were also developed; the "strip method" to measure in situ the interproximal, supragingival plaque-pH (Study I) and a microtiter plate format of RUT (Rapid Urease Test) to grade bacterial urease activity in vitro and ex vivo (Study III). Hopefully studies on pH and alkali formation in the dental plaque can contribute to finding measures for assessment of the individual caries risk.

The general aim of this thesis was to study the influence of various factors in posterior composite resin survival. A clinical point of view, beyond restorative materials' comparison, was the main objective of the investigations, focusing on the most frequent clinical failures, secondary caries and fracture. Specific concerns related to the material itself were also under evaluation.

The overall aim of this thesis was to study the influence of gaps and their size on the development of secondary caries.

Secondary caries development can be influenced by many factors: restorative or adhesive material used, loading of the restoration, micro- or macroleakage and width of the gap. The extent to which each of these factors contribute to secondary caries attack is undetermined, as is the possible interaction between them. Without this knowledge, it is impossible to establish rational clinical prevention against secondary caries or understand fully their etiology.
